"Is there anyone else?" ==> This is a neutral question. The person asking it does not have any particular expectation of whether the response will be affirmative or negative.

"Is there no one else?" ==> Although not completely sure what the response will be, the speaker has some reason to think or expect to be told "No, there is no one else." The speaker may also be hoping (but not really expecting) to be told that "There is someone else".
