The sentense should be :

"We had know each other for 3 year by April".

Sorry for confusing you.
Please let me know whether it is grammatically right and acceptable.

From what you have written it is not clear whether you are looking back to last April or looking forward to next April. There are therefore two possibilities:

By last April we had known each other for 3 years.

By next April we will/shall have known each other for 3 years.
