You could also say, " Where does John sit, do you know.?

Hi Suresh,

'Do you know where does John sit?' is incorrect because it's an indirect question, and in that case normal word order is applied to the following clause.

Direct question: Where does John sit?

Indirect question: Do you know where John sits?
