'Insist in' and 'Persist on' are the same meaning?? and they can be used as the same role? if not, how to use them?

Not really.

" I insist on your cleaning your room."
" You persist in not doing so."

To insist is to almost command.
To persist is to continue to ignore. (In the above example.)
