1. I want a small quantity of milk.
2. I want a little quantity of milk.
Which is the correct expression out of the above two?

'small' sounds more natural with 'quantity'.
However, you can replace 'quantity' with 'drop' if speaking about the amounts used in tea, coffee, etc. Then 'little' works too.
'I'd like a small drop of milk'
'I'd like a little drop of milk'
