1. Who of you can tell me what it is?
2. Who among you can tell me what it is?
Please correct the above questions if they contain mistakes.

In everyday English, the usual way to say this is "Which of you ..."

#1 sounds rather unnatural, though it is technically correct English in my opinion.

#2 is correct English but is more of a formal or literary form.

Dozy,
Is it colloquial to say:
"Which of you can tell me what it is?"

Yes.

[edited] By which I mean it is fine to use in everyday conversational English. However, it is not excluded from more formal English.

Is it right to say in this context:
'Can anyone tell me what it is?'

That is probably a far more common sentence than the examples above. It's fine.
