Why can't I use 'a' to have the meaning of a glass of water? I think I found a drink, a water can be used in some lesson. (I cannot remember which one.)

If you use 'a' to refer to the quantity or receptacle for the water, etc., then you have to state that clearly within the sentence.
a drink of water...
a glass of water...
a sip of water...
a litre of water...
'Water' itself is uncountable. It only becomes countable when the type of quantifier mentioned above is added.
Does your second sentence refer to having a drink of water? (Sometimes 'of' when said quickly sounds more like 'ah'.)
